The incorporation of O6-methyldeoxyguanosine monophosphate and O4-methyldeoxythymidine monophosphate into polynucleotide templates leads to errors during subsequent in vitro DNA synthesis

O4-MedTMP and O6-MedGMP may be incorporated from their 5'-triphosphates into polynucleotides by DNA polymerases. Once incorporated O6-MedGMP leads to errors during further cycles of DNA synthesis with a mutagenic efficiency of 0.3-0.7, the value depending upon the relative dNTP concentrations in the assay. In contrast O4-MedTMP leads to very few errors following its incorporation into DNA. The results demonstrate that O6-MedGMP, once incorporated into DNA, should be destined to give rise to DNA base changes during the second round of DNA synthesis.     

A benign chondroblastoma with a rare location

The benign chondroblastoma is rarely located at the leg and hand bones. The reported case had a deformation of the first phalanx of the left leg second finger, clinico-radiologically diagnosed as a chondroma, and only the histopathological examination sets the diagnosis of benign chondroblastoma.     

The phenomenon of the formation of universal rosette-forming cells under superextreme loads

A combined immunological study of cell-mediated and humoral immunity factors in sportsmen at the period of important competitions was made. The effect of the maximum bearable load was found to lead to the formation of a great number of universal rosette-forming lymphocytes and neutrophils. In the control group their number did not exceed 1-4% of the total population of these cells. Simultaneously, the suppression of the phagocytic activity of neutrophils and some of the humoral factors of systemic and local immunity with the parallel development of compensating processes were recorded.     

Prostaglandin modulation of the vascular effects of vasopressin in the conscious rat

Experiments were performed on conscious, male Sprague-Dawley rats to determine whether cyclooxygenase inhibition affects the pressor response to exogenous vasopressin. The rise in arterial blood pressure was tested in response to 1.0, 2.5, 5.0, and 12.5 mU synthetic arginine vasopressin both before and following cyclooxygenase inhibition with either meclofenamate or the structurally dissimilar inhibitor ibuprofen. In addition, time control experiments were also performed where only the saline vehicle for the drugs was administered. In all animals tested, the increase in arterial pressure in response to the highest three concentrations of vasopressin was greater following cyclooxygenase inhibition than before, while the saline vehicle had no effect. The baroreceptor-mediated bradycardia accompanying the rise in blood pressure was variable, but unaffected by meclofenamate or ibuprofen. It is concluded that vasodilator prostaglandins are released in response to pressor levels of vasopressin, which act to modulate the pressor response of the peptide.     

Vesicles and cisternae in the trans Golgi apparatus of human fibroblasts are acidic compartments

Recently we demonstrated that low-pH compartments can be visualized with the electron microscope using a basic congener of dinitrophenol, 3-(2,4-dinitroanilino)-3'-amino-N-methyldipropylamine (DAMP), which concentrates in acidic compartments and can be detected by immunocytochemistry with a monoclonal anti-dinitrophenol antibody. We now report that DAMP also accumulates in cisternae and vesicles associated with the trans face of the Golgi apparatus. DAMP rapidly leaves this compartment when cells are incubated with the ionophore monensin, which indicates that accumulation is due to the acidic pH in this compartment. Using indirect protein A-gold immunocytochemistry, we localized fibronectin, a major secretory protein in fibroblasts, to the trans Golgi vesicles that took up DAMP. Therefore, the trans cisternae of the Golgi apparatus and forming secretory vesicles have an acidic pH.     

How to become comfortable talking about sex to your patients.

Dealing with patients'' sex-related complaints is uncomfortable for many physicians. It can become more comfortable if the physician learns to apply his or her clinical knowledge of sexuality in combination with specific interpersonal interviewing skills. The level of comfort can be increased if the physician accepts his or her professional limitations in providing therapy and knows what other sources of care are available.     

Group B streptococcal septicemia and delayed-onset congenital right-sided diaphragmatic hernia.

A case is reported of fulminant early-onset group B streptococcal septicemia and delayed-onset congenital right-sided diaphragmatic hernia in a neonate. The latter condition should be considered when early-onset group B streptococcal disease is followed by increasing respiratory distress, right-sided pleural effusion and partial or complete opacification of the right side of the thorax.     

Protection of crystal-induced polymorphonuclear leukocyte membranolysis by phosphocitrate

The protection afforded by phosphocitrate, a phosphorylated polycarboxylic acid, against crystal-induced membrane damage to polymorphonuclear leukocytes was studied in vitro. Membranolysis was assessed by nitro blue tetrazolium salt reduction, lactate dehydrogenase release, and scanning electron microscopy. Phosphocitrate protected strongly against hydroxyapatite crystal-induced damage, an action attributable to crystal surface binding of phosphocitrate rather than to the membrane. The ability of phosphocitrate to prevent hydroxyapatite crystallization, together with its membrane protective effect against preformed crystals, would suggest that the compound might have a useful future role against crystal-induced arthropathies.
